Innovating Antibody-Oligonucleotide Conjugates (AOCs) for Enhanced BBB Penetrance
With BBB-penetrating technologies at the forefront of Korean innovation, the next frontier lies in moving beyond antibody shuttles to explore receptor‑mediated transcytosis for delivering therapeutic oligonucleotides to the brain. In this interactive workshop, explore the possibilities of platforms such as ABL Bio’s Grabody-B being utilized to shuttle ASOs and siRNAs across the BBB, and ponder questions such as:
- What do we understand about the behaviour of oligonucleotides when conjugated to antibodies and how can we optimize the conjugation for effective delivery?
- How can we overcome the challenge of siRNA negative charge limiting intracellular delivery in the CNS?
- How can we maximize endosomal escape of IV-administered oligonucleotides in the CNS?
- What are the advantages of delivering oligonucleotides via RMT vs intrathecal administration?
